چکیده

1. Besides miR-133a, miR-133b and miR-208a, there are several other muscle-specific miRNAs, such as miR-1, etc. Why only these miRNAs were used in this study? 2. P2: qRT-PCR is not quantitative real-time polymerase chain reaction, but quantitative reverse transcription-polymerase chain reaction. 3. P6: “To date, no housekeeping miRNAs have been established and validated to normalize for the miRNAs content.” It’s wrong. Actually, miR-16 has been used as a reference in the detection of body fluid miRNAs (Cui L, et al. Cancer. 2013 May 1;119(9):1618-26). Should gave some discussion about this.

Circulating miRNAs reflect early myocardial injury and recovery after heart transplantation

BACKGROUND MicroRNAs (miRNAs) are short, single-stranded and non-coding RNAs, freely circulating in human plasma and correlating with vary pathologies. In this study, we monitored early myocardial injury and recovery after heart transplantation by detecting levels of circulating muscle-specific miR-133a, miR-133b and miR-208a. METHODS 7 consecutive patients underwent heart transplantation in ...

Heart Failure Loss of AngiomiR-126 and 130a in Angiogenic Early Outgrowth Cells From Patients With Chronic Heart Failure Role for Impaired In Vivo Neovascularization and Cardiac Repair Capacity

Background—MicroRNAs are key regulators of angiogenic processes. Administration of angiogenic early outgrowth cells (EOCs) or CD34 ϩ cells has been suggested to improve cardiac function after ischemic injury, in particular by promoting neovascularization. The present study therefore examines regulation of angiomiRs, microRNAs involved in angiogen-esis, in angiogenic EOCs and circulating CD34 ϩ ...

Plasma miR-208 as a biomarker of myocardial injury.

BACKGROUND MicroRNAs (miRNAs) are endogenous small RNAs of 21-25 nucleotides that can pair with sites in 3' untranslated regions in mRNAs of protein-coding genes to downregulate their expression. Recently, circulating miRNAs have been reported as promising biomarkers for various pathologic conditions. We assessed the hypothesis that miRNAs may leak into the circulating blood from injured cells ...

Use of diffusion tensor imaging to predict myocardial viability after warm global ischemia: possible avenue for use of non-beating donor hearts.

BACKGROUND The assessment of myocardial viability after global warm ischemia (WI) but before reperfusion is challenging. We hypothesized that fractional anisotropy (FA), a magnetic resonance imaging (MRI) parameter of water diffusion that characterizes cellular integrity within tissues, provides a rapid and useful method for evaluating the viability of hearts after WI. METHODS Dog hearts were...

Targeting microRNAs to promote cardiac repair and cardiomyocyte proliferation as a potential regenerative therapeutic approach

MicroRNAs (miRNAs) have emerged as important modulators of cardiovascular biology, repair, and regeneration. miRNAs posttranscriptionally regulate the expression of a network of proteins and consequently, cardiovascular development. miRNA targeting has been shown to: (i) improve the cardiac repair capacity of patient-derived circulating or bone marrow–derived mononuclear cells (BMCs); (ii) indu...
